Hans Christian Gram received a Doctor of Medicine degree from Copenhagen University in the year 1878. After this, he went to Europe to study bacteriology and pharmacology. While working at the Microbiojilt Lab in Berlin, he notices that mixing of bacteria spots into crystal violet stains (iodine solution and organic solvent) gives different structure and biochemical function in different samples.

Hans Christian Gramm published his discovery in a journal in 1884 under the names Gram-positive and Gram-negative.
